1 – Principles of Biochemistry

late 1800 ได้มีการพัฒนาchemist ในยุโรป เพราะต้องการหาวิธีทำ alcoholic fermentation เพื่อ improve quantity and quality of beer , wine
1897 German chemist Eduard Buchner ค้นพบวิธี extract yeast cell in vitro , เพื่อ convert glucose -> carbon dioxide , ethanol ภายใต้ anaerobic condition
ค้นพบ some yeast protein function เป็น chemical catalyst

1970 technique develop manipulate deoxyribonucleic acid (DNA ) เป็น recombinate DNA technology , สร้าง human insulin bacteria

Understanding molecular level
1. hierarchical organization biochemeical process within cell , organism ,ecosystem
2. DNA เป็น chemcial basis heredity and encode structual information for RNA and protein molecules – เป็น mediated biochemial process ใน cell
3. function of biomolecule ถูก determine โดย molecular structure , fine-tuned ผ่านทาง random DNA mutation and natural selection

history – Eyptians 2000ปี สร้าง intoxicating substance (ethanol ) and caustic acid(acetic acid)
Greek ใช้ “zyme” หรือ yeast เพื่อสร้าง gas (carbon dioxide) ในขนมปังและ grapes into wine
Eduard Buchner 1897 ทำ “fermented of zyme” หรือ enzymes , in vitro reaction
– ใช้ mixed diatomaceous earth(kieselguhr) , เพื่อ prepare , extract
– ถือว่าดี เพราะหลีกเลี่ยงการทำ extract alkaline and inactive ที่เกิดเมื่อ yeast protein contact with glass
– จากนั้น ทำหลายวิธีเพื่อ preserve to prevent coagulation , เขาใช้ 40% sucrose solution เพื่อ glucose for alcoholic fermentation
– จากนั้น ได้ใช้ stain yeast – Saccharomyces cerevisiae เพื่อทำ local brewery in Munich ซึ่งเป็น strin ที่ดีกว่าใน paris

All living cellต้องมี enzyme , ทั้ง protein , RNA function , reaction catalysts เพื่อเพิ่ม rate biochemical reaction
